Is it possible to play sound files on an Android smartphone directly from Home Assistant? Does the sound file need to be present on the smartphone for this to work?
Could you provide an example for my specific use case?

I'm currently using an alarm notification with a custom ringtone that I uploaded to my Android device.
Did you try looking at the docs?
Check Supported media types - there is only iOS support
